We have our application currently sitting on a t3.medium instance (2 CPU, 4GB RAM) with an RDS database. We have 4 workers configured. However, the system is not as fast as expected. Hence, we want to upgrade. Which instance size should we go for to support up to 400 users (20% of which will be concurrent)?

Is that really possible? I have users in a single location using ohio on aws and its crashing using t3x.large and I only have 10 users? how is that possible?
I think you need to upgrade to a large instance.
I have 170 databases with users on 1 single t3.medium instance and it's working fine.
Make sure that select a near physical regional location
