How to set created Template as the Default template used to send mail automatically ?

I'm gonna assume you are talking about the sales quotation. Enable the developer mode, go to the sales quotation, click "Send by Email", select the template you want as default and then from the dropdown in the upper left corner, select "Set Defaults", choose "Template = My template whatever it's called" from the Default dropdown, select if you want to apply it for yourself only or for all users and click "Save default".
