How do I send email to employee/applicant when leave is refuse by an office or manager in odoo 12 CE.

Use odoo Chatter area to send mails.
Or Create an automated Action
1) Enable Developer mode(Go to settings-> Activate the developer mode
2) Go to Setting->Automated Actions
3) Create a new action
Action Name - > Leave Refused
Model -> Time Off
Action To Do -> Send Email
Trigger Action -> On Update
Apply on -> Status = Refuse
You can define the groups too
