Can I remove the "To Read" filter as the default filter when viewing To-do? That way I would see all of my starred messages there by default. I tried activating developer mode but "To Read" is not listed under Manage Filters.

You need to remove the context value 'search_default_filter' from the action of that menu, where filter is the name of the filter
Can that be done via the UI? If so, where? If not, which file should be edited?
Yes. Just activate the developer mode in the admin user, edit the action and change the value of the field context
