From quotation i sent an email to the customer , so he can sign and approve it. But if the customer is not a portal user , when he access the link system shows a NOT FOUND error. It is require to have the customer as a portal user in order to give him the access to sign documents ?

You have to grant the portal user from the customer master itselt. Then only the can access the links. eg:
Use Grant Portel Access here for the related customer.
