I have an instance running odoo in a free instance of amzon , I wanted to map my URL to that ip where the odoo is running.
Is Ngnix is required and how to configure from scratch

You can use nginx or any other proxy server like Apache, Caddy Webserver etc. for that purpose. Google does provide plenty of good readings for the setup.
